If you have an unused OEM DVD of Windows 10 Pro 64 pit that you purchased form a retail store, can you use the product key from the original release version on the DVD with a clean USB install of 1607 made from a downloaded ISO from Microsoft?
Or will the product key not be recognized as the 1607 ISO is retail channel or will the activation juts fail?
I hate to go through a reinstall process or and upgrade process just to get to 1607 from 10240.16384 or 10586....

It should work, you can simply test it by booting up the 14393 Multiple Editions iso and enter the key at initial setup.

Yes you can, I have done it myself with Win10 Home OEM. You can also use windows7/8/8.1 product key and it will be activated upon install. Seems they are still honoring the free upgrade in some respects.

Frankly in 30+ years of IT i never touched a retail windows installation personally. No one minds to use them

    Corporate have dedicate offers.

Single users are divided in pirated copy, bundled OEM copy, and standalone OEM

Probably there are few fools who spent 500$ to get Vista ultimate, but they are a tiny minority

Yes. I downloaded an .iso from the same link about a week after the Anniversary Edition was released and that's exactly what I got.

The current Iso from MS does not have the Cumulative updates. The last 1607 Cumulative update has caused pin and password problems on five different machines I have encounter, not any of may machines, but if you enter the pin or password it cycles (swirling ants and then it comes back to entry box). Machine must be shut off for some time and in some cases the cmos battery removed to get it to come back. OEM key does in fact work with newest iso verified with Pro. We have called MS and they have not and will not acknowledge password pin issue with the latest cumukative for 1607. We were told to reload a laptop with this problem , that they could not fic over the phone. We did a reload with anniversary issue and after cumulative updates, & pin issue was back, Acer I5 laptop. Uninstalling or rolling back fixes until it is reinstalled. If date and time do not show on lock screen, upon first boot no entry. If they are present machine takes pin. If they are present after first boot, and pin causes re-cycle to lock, no acceptation of pin, till turned off for some time. I am not sure of problem is only with UEFI boot and not MBR.
