    In a PC there are 4 NTFS partitions C D E and F.
    By diskpart following have done:

    diskpart: select volume 3
    diskpart: remove letter E
    diskpart: select volume 4
    diskpart: remove letter F

Now in my computer only C and D drives are appearing.

Q1 can any program still have access on drive E or F.

Q2 If an antivirus run to scan full computer e.g. Microsoft Security Essentials then will it scan drive E or F or only restricted to drive C and D.

You have only removed the letter I still think Microsoft Security Essentials will scan E and F. Easy answer is to try running Microsoft Security Essentials and see if it detects the drives. What are you really trying to achieve, also if you go into disk management you will probably see those drives.
