Just format the usb thumbdrive FAT32 using explorer and extract the iso to it, done. Now you have an UEFI & Legacy BIOS bootable USB Thumbdrive without using any 3rd party software.
That surely works. But still rufus comes handy because it can do it with tho mouse clicks, and can do that not only starting from Vista-W10 ISOs but also XO, win2k, most linux and bsd distros. And can do that even if your actual system is not w10.
Is this something new with Windows 10? I know when I only formatted my USB thumb drive and copied over the .iso files it was not bootable. You sure you can do this with Win 10?
It's deadly slow compared to rufus. And unlike rufus that requires just a click, it needs to be installed. Do a favor to yourself. Just use rufus.

Ensure the registry key "HKCU\SOFTWARE\Microsoft\ISO Backup Tool" is created. 2. Create a new DWORD value named "DisableFormat" in this location and set the value to 1. NOTE: The USB device should be formatted manually before running the tool. if you format to fat32 before running the tool (using the above reg entry), it will. Rufus FTW though
I use Easy2Boot. I have a 16g flash drive that has both 32 and 64bit Win10 on it (from MS Media Creator so they have both the Home and Pro built in) as well as Win7 All-In-Ones for 32/64bit and Parted Magic. Works great for me and was very easy to setup. T
I testing WinToHDD right now on an extra Internal HDD (instead of DVD Drive) on my Laptop. Procedure is easy, if have the Apps installed and a Windows 10 ISO ready (which I did): Prepare HDD with partitioning to: Partition 1 System Reserved (500MB) and Partition 2 Boot Partition, sized to your need Start WinToHDD chose the .ISO file choose the HDD start the process! The creation of the first part is done within 3min!! Restart the computer and boot from that newly installed HDD. Computer will restart again after about 2min! Initial setup of Hardware (driver etc.), download of critical Updates (depend on used Version), and personal, done in about 5-7min. I use en_windows_10_enterprise_2015_ltsb_n_x64_dvd_6848316.iso and the whole procedure of install and setup, incl. local account etc., etc., just took 25min, a bit more short that a 'normal' clean install as it normally take, on that 5 years old Laptop! I activated with using MKTV26B5, with no problems. Main advantage of that Apps WinToHDD, seems to be that it would be done with an available internal Windows HDD/System, without to interfere with that in any means. Just use the BIOS Boot Manager while start the machine, to choose what HDD/OS to start. Also, after initial install (step 1) of Windows, and before starting the Settings setup (after the first Restart), that HDD could be taken into any other Machine and finalize there! A good solution for System Builders and/or Computer Retailers!

The MCT is meant for downloading the wu esd files and creating either an ISO or directly a bootable USB drive. The win 7 store usb/dvd tool can be used to create a bootable usb drive or burn it directly to dvd from other iso (legacy bios only). Or format a usb thumbdrive FAT32 and extract the iso to it, to create a Legacy BIOS and UEFI bootable usb drive.
